Knowing that Vent and other voice chatting will most likely going to be used while playing LoL I think the Audio settings may need to be a bit more advanced. Mainly choosing your Input and Output of the sound.

I have a headset that, when plugged in, directs all sound to it. This is easily fixable by changing in-game sound output which LoL currently lacks. Personally my headset's headphones don't work and only the mic does, so no sound for me! Although that is just a personal problem I still think the Audio settings sound have more advanced capabilities.

Set your default audio playback device in Windows to your speakers and in Ventrilo settings set it to your headset. Do this while the headset is plugged in. This way you can hear your team without it being drowned out by in-game sounds.
