You NEED a jungler for sure if they know how to jungle right the 2 in the lane will be higher lvl than all of the other team (unless they have a jungler also) then you need that most heavy health dps you can get for example my old team ran kennen mord nunu, nunu interchanges a lot with ww and brolaf but they lost maybe 6 times out of 100+ games maybe? we also ran tryn malphite and nunu

Designated Jungling doesn't really work because simply put, there isn't enough Jungle nor flow within the jungle to do it properly. Putting 1 talent into Smite is typically a waste in a map like Twisted Treeline. Specifically, the Red buff really only helps your assassins, and they should be roaming either way.

You're better off either evening the bottom fight out 2v2, or if they have one person jungling, your 2 guys should have no issues pushing bottom back to their turret, then both slide up top to Gank top 3v1, or slide in and counter Jungle.

Nunu is a strong player because his slows and damage are good, Morde is good simply because of his ability to survive most fights. I'd probably roll with Trynd over Kennen because of Trynd's superior Mobility on Twisted Treeline. While Kennen is nice and fast, Trynd has a free Flash over the wall which is quite useful, and twisted Treeline, specifically around Dragon and Red Buff, has these walls that take a long time to go around, so Trynd's spin really is a spin to win.

Both of those teams are really bad Combos, specifically because they are entirely Physical Damage oriented. While Janna does Magic Damage, she's meant as a support character and doesn't really have the nuke required to down people. If you roll with either of those teams, people are going to get Thornmail and your Melee champs are going to have a rough time. Especially as Kat and Lee Sin are both Classified as Assassins, they'll be a bit squishy, and your team will have no counter to a good Alistar, Malphite.

As a rule of thumb, aim for 1 Magic, 1 Physical, and 1 Ranged. Typically a Mage, Assassin/Bruiser/Tank, and Ranged AD/Another Mage.

And the More CC the better - because come mid game if both teams have lost towers, the amount of space between bases seems huge, and being able to escape or keep enemies from escaping is unparalleled. But thats also why Trynd is so power heavy on this map, he has a lot of escape routes that people can't always follow.
