I've seen great success with a lot of combos, mostly depending on their teamwork and skill, but there are a few that stand out:

Blitzcrank+anyone=a tower hook and stun means death, no matter who your partner is (though another stunner tends to work best with this).

Blitzcrank + morgana=no escape. Long disable, lava, stun, hook, more stun (blitz's cooldown is very low), double ult, and nobody escapes even halfway through the combo. They also are pushing monsters.

Blitzcrank +Singe=Conceptually weird (two pusher tanks 0_0? Don't they get harassed to death?), but works. Hilarity ensues with hook, stun, poison trail, slowing goo, more blitz stun, fling (if they are still alive), and even more blitz stun. Plus, blitz and singe push like beasts.

Amumu/Nassus/anyone with a ranged, reliable slow+Ryze=nothing like a good slow to let Ryze catch up, prison, and ult. They lack pushing power, but why push when you can kill and then push? After a few combos like this, players won't even come near the minions, for fear of being killed.

Chronokeeper/Taric+Yi/Jax/Evelyn=while not exactly a killer combo, Chronokeeper's harassment bombs to keep enemies away, slow to save allies, and revive keep melee carries/nukers in the fray for as long as possible, helping buff up their late game considerably. Taric helps out with healing, a ranged stun, and extra armor. Chronokeeper pushes with his harassment bombs, and Taric has his AOE spell and an ult to push.

Pirate and Dr. Mundo=Weird-sounding at first, but any ranged hero suddenly losing tons of hp from parley and cleaver, along with a reliable slow (cleaver) to let the pirate catch up and fire more parleys usually kills or at least puts down any ranged hero, much less a melee hero. Plus, both are hard to harass from pirate's heal and Mundo's natural regen abiliy. Not exactly the best pushers (at least early game), but nobody can come within 500 units of the minions without taking serious damage, leaving little to defend against.

Singe+Ryze=Good times. Fling, prison, Ryze ults. What else is there to life? Singe pushes in this case.

TEAMWORK=The most important part of laning. Any combo can be godlike with good communication and teamwork. Talk with your lane partner and work out tactics. Smooth cooperation and good reactions are the only reason any combo works.

That's all I have for now, maybe I'll add more later. If anyone wants to correct me, please feel free, but please no flaming/swearing/"you are a ****** and you suck" comments without decent justification. Constructive criticism only please.

Also, Sivir and Mundo. Mundo tanks and slows, while Sivir with Stark's (and what Sivir doesn't get Stark's?) gives Mundo everything he needs - attack speed, lifesteal... And with 3-4 levels in Burning Agony, they can push lanes so friggin' fast.

Me and a friend of mine duo quite often in set pairs. The ones we've tried and liked are;

Twitch/Gangplank. We like the two poison passives, which is among the reasons we started. Gangplank often uses parrrlay for minions early on but occassionally harasses heros. Often we let gangplank get lowish on HP to coax greed kill behavior, then have them chase the pirate (as he pops a parrrlay when he can) and twitch sneaks up to make a kill.

Also we like.

Rammus/Master Yi. Dunno if this is particularly potent, but Rammus can tank and Yi DPS. Generally we'll try and get close enough to do Piercing Taunt (Esp near a tower) to get someone on Rammus while Yi lets out on them, when taunt is over we power ball and pin. Not to mention they both have excellent anti-tower capabilities.

We theorized that Nasus/Morgana would be good but it seems they arn't our types of characters unfortunately. But the above two pairs we tend to do very well with, in many games getting the lions share of kills.

Anivia/Ashe works brilliantly. Once level 6 rolls around its pretty much an instant kill. Lead with either an Enchanted Arrow or a Flash Frost, then Frost bite, then do the other for the kill. Here's how it breaks down.

(Assuming you're in the bottom or top lanes), FF (Flash Frost) out of the bushes, get that mini-stun. From there Ashe can then fire his arrow at the immobile target for an easy shot, while at the same time Anivia throws out her first Frost Bite. If the hero is not dead yet, put the edge of the Glacial Storm on the hero, while the rest is towards his retreating area. (this forces him to run through it, or have to take more time to run around). Anivia then follows while Ashe chases with Frost Arrows and volley and finishes with a Frost Bite.

This would at least force a Flash to escape or net you a kill. Some acceptions may vary, Morganna and Kayle being two, but aside from that its almost a sure kill with the damage you can crank out.

Note: this works well for advanced and beginner play as you can switch the order to land the Arrow first (much easier to than the slower moving Flash Frost) and follow up with FF. This option may be optimal for Anivia players who are still getting the FF timing down, or simply are lagging a bit too much.

Morgana + Annie. Easiest duo to first blood with if you both know the champions well.

A great combo is Morgana + Veigar - Level 1 Dark Binding and Lvl 1 Dark matter... the binding lasts long enough at level 1 to hold them so dark binding hits. So whenever Morg is able to root one... you got a huge amount of dmg done.

These two can also push well with their AOE
